Saint Marinus, who fled his country in Dalmatia (now Croatia) during the Diocletianic Persecution of Christians, founded the historic Republic of San Marino, which bears his name. Marinus sought sanctuary on the Italian peninsula at Monte Titano, where he erected a chapel and established a monastic community in 301 A.D. San Marino was founded on the location of a community of Tongva (Gabrieleo) Indians who lived near where the Huntington School now stands. The land was part of the San Gabriel Mission's holdings. (The Mission's grist mill was known as the "Old Mill.") Major portions of San Marino were included in a Mexican land grant of 128 acres to Victoria Bartolmea Reid, a Gabrielea Indian, in 1838. (She wedded Scotsman Hugo Reid in 1837 after her first husband, also a Gabrieleo, died of smallpox in 1836.)

Rancho Huerta de Cuati was the name she gave to the property. Seora Reid sold her rancho to Don Benito Wilson, the first Anglo owner of Rancho San Pascual, after Hugo Reid died in 1852. Don Benito gave 500 acres (2.0 km2) to his son-in-law, James DeBarth Shorb, in 1873, including Rancho Huerta de Cuati, which Shorb dubbed "San Marino" after his grandfather's estate in Maryland, which was named after the Republic of San Marino on the Italian Peninsula in Europe.

The city, which is nestled in the San Rafael Hills, is divided into seven zones based on minimum lot sizes. The smallest lot is roughly 4,500 square feet (420 m2), although many of them are larger than 30,000 square feet (2,800 m2). Because of this and other causes, most of the homes in San Marino, which were built between 1920 and 1950, do not look like those in nearby Southern California neighborhoods (with the exception, perhaps, of neighboring portions of Pasadena). San Marino's residents have also developed a spirit of historic preservation. The city's strict design review and zoning restrictions have so far prevented the development of big residences found elsewhere in Los Angeles, with small exceptions.
